Successful and popular comedian George Simmons (played by Adam Sandler) discovers he has leukemia and decides to return to the stand-up comedy stage while he still can. During a performance, George meets aspiring stand-up comedian Ira (played by Seth Rogen). Struggling financially, Ira has always dreamed of becoming a professional stand-up comedian, and he is currently staying in the living room of his friend, TV actor Mark (played by Jason Schwartzman), due to his inability to pay rent. Also living there is fellow stand-up enthusiast Leo (played by Jonah Hill). Ira is hired by George as a personal assistant, convincing him to share his illness with his family and ex-fiancée Laura (played by Leslie Mann). George's leukemia is brought under control with a new experimental drug, and he excitedly travels to San Francisco with Ira to perform, where old feelings reignite at Laura's home in Marin County. However, just as things start to heat up, Laura's husband Clark (played by Eric Bana) unexpectedly returns home, making everything awkward and disastrous...
