The story is set in a future timeline where the rampant dog flu prompts the mayor (voiced by Kunichi Nomura) to enact a dog exile law, banishing all dogs from the city to "Trash Island" to fend for themselves. The mayor's guard dog, Spots (voiced by Liev Schreiber), is the first to be affected. Thus, the once pampered dogs that lived in homes now find themselves scavenging for food in piles of garbage. A group of five stray dogs on the island—Leader (voiced by Bryan Cranston), Duke (voiced by Edward Norton), Boss (voiced by Bill Murray), King (voiced by Bob Balaban), and Chief (voiced by Jeff Goldblum)—typically stick together. One day, the mayor's ward, Atari (voiced by Koyu Rankin), suddenly lands on the island in a plane, seeking his lifelong friend, Spots.
